Its role extends beyond biological considerations by supporting formulation robustness through protection of oxidation-sensitive lipid components during storage and routine clinical use.
Unlike Regional Signature Components, mixed tocopherols are incorporated throughout the platform to provide a common antioxidant foundation and to enhance long-term formulation stability. Their inclusion contributes to platform consistency rather than regional differentiation.
Within the Release–Integration architecture, Vitamin E is regarded as a universal support component integrated into the common formulation backbone.
Scientific rationale
Vitamin E comprises a family of naturally occurring lipid-soluble antioxidants, including four tocopherols (α-, β-, γ- and δ-) and four tocotrienols. Mixed natural tocopherols provide a broader antioxidant profile than isolated α-tocopherol because each homologue exhibits distinct chemical properties and preferential interactions with different lipid substrates.
Experimental studies consistently demonstrate that vitamin E protects membrane lipids against oxidative degradation and contributes to maintenance of epidermal barrier integrity.
Topically applied vitamin E has been investigated in numerous dermatological studies evaluating skin hydration, barrier support, photoprotection and oxidative stress. Although clinical findings vary depending on formulation and study design, its favourable safety profile and antioxidant properties are well established.
Within pharmaceutical formulations, mixed tocopherols are also widely employed as natural antioxidants to reduce oxidative degradation of plant oils and other oxidation-sensitive ingredients, thereby improving product stability.
Current scientific evidence primarily evaluates topical vitamin E within conventional dermatological preparations. At present, no published studies specifically investigate mixed tocopherols as components of region-specific neurodermal formulation systems such as the SIRIAM Platform. Their incorporation therefore reflects both established pharmaceutical practice and the design philosophy of the platform rather than evidence for region-specific application.

Its primary architectural role is to stabilise the lipid environment of the formulation while contributing to dermal compatibility. Rather than defining regional identity, it reinforces the pharmaceutical consistency shared by multiple magistral preparations.

Current platform development supports continued use of Full Spectrum Natural Vitamin E as the preferred antioxidant system.
Future optimisation may evaluate:
concentration refinement according to oil composition;
compatibility with emerging botanical ingredients;
long-term oxidative stability under accelerated storage conditions;
interaction with copaiba oil, castor oil and squalane;
preservation of antioxidant activity throughout shelf life.
Replacement by isolated synthetic α-tocopherol is not currently envisaged, as the broader antioxidant profile of mixed natural tocopherols better aligns with the SIRIAM formulation philosophy.
